The hidden blade has no counter animation throwing the stronger opponents to the ground, so when you do a counter attack w/ your hidden blade (it is hard, but it is possible) then you have a instand kill. It even works on Targets. I have killed Target number 10 w/ this technique, but it felt like I missed something...
After you beat the game, you can continue playing by walking into your room, and then use the new gift. Then after the reeeeaaaaly long credits, you can load a memory block in the Animus (what is latin and means 'heart' by the way).

After the credits roll at the end of the game you'll get an access code for the computer attached to the Animus. Once you hack the computer, you'll see an inbox, outbox and deleted message. Look at the inbox and choose the entry regarding the conference room. Now you can go to the conference room and access it via the keypad.
After beating the game, you can use Eagle Vision inside the Animus at any time, regardless of your health level (Synchronization).
After beating the game, you can kill anyone inside the Animus at any time, without losing health (Synchronization).
The best weapon against multiple gaurds is the shortblade.
